Best Transportation Companies To Invest In Right Now: Juno Therapeutics Inc (JUNO)

Juno Therapeutics, Inc. (Juno) is a United States-based clinical-stage biopharmaceutical company. The Company is engaged in developing cell-based cancer immunotherapies based on its chimeric antigen receptor (CAR), and high-affinity T cell receptor (TCR), technologies to genetically engineer T cells to recognize and kill cancer cells. Through genetic engineering, the Company inserts a gene for a particular CAR or TCR construct into the T cell that enables it to better recognize cancer cells. Its CAR technology directs T cells to recognize cancer cells based on the expression of specific proteins located on the cell surface, whereas its TCR technology provides the T cells with a specific T cell receptor to recognize protein fragments derived from either the surface or inside the cell.

To provide treatment, the Company harvests blood cells from a cancer patient, separate the appropriate T cells, activate the cells, insert the gene sequence for the CAR or TCR construct into the cellsDeoxyribonucleic Acid (DNA), and grow these modified T cells to the desired dose level. The modified T cells can then be infused into the patient or frozen and stored for later infusion. Once infused, the T cells are designed to multiply, through a process known as cell expansion, when they encounter the targeted proteins and to kill the targeted cancer cells.

CD19-Directed Product Candidates

The Company product candidates, such as JCAR015, JCAR017, and JCAR014, which use CAR technology to target CD19, a protein expressed on the surface of almost all B cell leukemias and lymphomas. The Company has rights to commercialize each of these product candidates across the world. The Company JCAR015 is an advanced development product candidate, and it has demonstrated clinically meaningful complete remission rates in adult patients with r/r ALL. JCAR015 uses the CD28 costimulatory domain and CD3 enriched peripheral blood mononuclear cells (PBMC), which requires fewer process steps for! manufacture than its defined cell composition product candidates. JCAR015 is being tested in a Phase I open label clinical trial of patients with r/r ALL for the treatment of refractory chronic lymphocytic leukemia.

JCAR017 also targets CD19, but differs from JCAR015 in several important respects. JCAR017 uses the 4-1BB costimulatory domain and a defined cell composition of CD4+ T cells and CD8+ T cells. JCAR017 is in development for pediatric patients with r/r ALL. JCAR017 is being evaluated in a Phase I/II clinical trial in pediatric r/r ALL.

JCAR014 also targets CD19. JCAR014 uses the 4-1BB costimulatory domain and is composed of CD8+ central memory T cells and CD4+ T cells in a defined ratio. JCAR014 is being evaluated in a Phase I/II trial as a treatment for a number of B cell malignancies in patients relapsed or refractory to standard therapies.

Additional Product Candidates

The Company is exploring the potential of its CAR and TCR technologies against targets that have the potential to treat cancers not targeted by CD19-directed products, in particular, difficult-to-treat solid organ tumors, such as certain breast, lung, and pancreatic cancers, as well as B cell malignancies that do not express CD19. The Company CD22 is a cell surface protein widely expressed on B lymphocytes. It is expressed by B cell malignancies, including non-Hodgkin lymphoma (NHL), acute lymphoblastic leukemia (ALL), and chronic lymphocytic leukemia (CLL). The Company L1CAM, also known as CD171, is a cell-surface adhesion molecule that plays an important role in the development of a normal nervous system. It is overexpressed in neuroblastoma, and there is increasing evidence of aberrant expression in a variety of solid organ tumors, including glioblastoma and lung, pancreatic, and ovarian cancers.

The Company MUC-16 is a protein overexpressed in the ovarian cancers, but not on the surface of normal ovary cells. CA-125 is a protein found in the ! blood of ! ovarian cancer patients that results from the cleavage of MUC-16. CA-125 levels in the blood are a common test for ovarian cancer progression because they correlate with cancer progression. Its MUC-16/IL-12 product candidate uses its armored CAR technology. IL-12 is a cytokine that can help overcome the inhibitory effects that the tumor micro-environment can have on T cell activity.

The Company ROR-1 is a protein expressed in the formation of embryos, but in normal adult cells its surface expression is predominantly found at low levels on adipocytes, or fat cells, and briefly on precursors to B cells, or pre-B cells, during normal B cell maturation. ROR-1 is overexpressed on a variety of cancers including a subset of non-small cell lung cancer, triple negative breast cancer, pancreatic cancer, and prostate cancer. It is expressed on B cell chronic lymphocytic leukemia and mantle cell lymphoma. The Company high-affinity TCR T cell product candidate targets WT-1, an intracellular protein that is overexpressed in a number of cancers, including adult myeloid leukemia, or AML, and non-small cell lung, breast, pancreatic, ovarian, and colorectal cancers.

The decliners keep greed in check while reminding us about the risks of the equity markets. Let's go over some of last week's best and worst performers. Regulus Therapeutics (RGLS) -- Up 161 percent last week Last week's biggest winner was a biotech upstart that got welcome news on a potential hepatitis C treatment. Regulus Therapeutics is showing that patients receiving a single injection of its drug are reporting lower viral loads a month later. There are still plenty of regulatory hurdles to clear, but it's certainly encouraging. Select Comfort (SCSS) -- Up 21 percent last week Shares of Select Comfort moved higher after it announced blowout quarterly results. The company behind the Sleep Number air-chambered mattresses saw net sales and earnings per share climb 23 percent and 22 percent, respectively. Select Comfort's strong performance was fueled by an impressive 16 percent spike in comparable-store sales. The favorable momentum is going to linger: Select Comfort is boosting its guidance for the entire year. Six Flags (SIX) -- Up 16 percent last week It was a summer of thrills at Six Flags. Shares of the regional amusement park operator rose like a coaster on a chain lift after it announced better-than-expected revenue growth. Revenue increased 7 percent on a combination of a slight uptick in attendance, higher admission prices, and guests spending more once inside the park. The report was encouraging enough for Six Flags to boost its dividend. The stock is now yielding a hearty 5.3 percent. Boulder Brands (BDBD) -- Down 31 percent last week Shares of Boulder Brands lost nearly a third of their value after the food company behind Smart Balance buttery spreads and EVOL frozen entrees warned of a soft holiday quarter. It now foresees net sales of $132 million to $137 million for the fourth quarter. We would be looking at a sequential dip from

Similarly, a delivery service in the states could be a novel idea if Burger King Worldwide (NYSE: BKW  ) hadn't already beaten McDonald's to the punch. In early 2012, Burger King began testing a delivery service in Washington D.C., using specialized thermal packaging that keeps hot food hot, and cold drinks cold, during the delivery process. Allowing consumers to order nearly the full gambit of items you can get in the restaurant and placing a minimum order price to ensure margins are met has allowed Burger King to one-up its peers. Since this pilot program, Burger King has expanded it to Houston, Miami, and New York, and announced just last week that Los Angeles, Chicago, and San Francisco are up next.

This is where we look out
That's a good reason to be worried, if you're an investor in Starbucks (NASDAQ: SBUX  ) or Green Mountain Coffee Roasters (NASDAQ: GMCR  ) or really any major coffee company. Master Blenders' main line is a brand called Douwe Egberts, which used to be served in Burger King (NYSE: BKW  ) until the company signed with Seattle's Best, a Starbucks brand.

  • [By Sean Williams]

The restaurant sector is extremely competitive to begin with, so any negative publicity can crush a company, regardless of its size or dominance, over the short term. This week has been something of a nightmare for fast-food restaurant chains McDonald's (NYSE: MCD  ) and Burger King Worldwide (NYSE: BKW  ) .

  • [By WWW.DAILYFINANCE.COM]

Andrew Harrer/Bloomberg via Getty Images It's apparently never too early to have a hamburger. While most chains wait until 10 or 11 a.m. to begin offering their signature burgers, early risers will be able to skip straight to lunch at thousands of participating Burger King (BKW) locations. The fast food giant's betting that some morning commuters want more than just a breakfast burrito or a ham and egg sandwich for breakfast. Later this month Burger King is rolling out a "burgers for breakfast" campaign across 5,000 of its restaurants, offering its signature Whopper as well as a few other burgers and chicken sandwiches in the morning. Assuming that you don't want hash browns on the side, fries will also be made available. So will apple pie. Whopper, BK King and Original Fried Chicken sandwiches will be part of the chain's breakfast offerings. Will it confuse diners? Will the move complicate matters for employees manning the prep tables and fryers? Will the great variety stall patrons as they ponder the menu when it's time to order, creating longer wait times for food? The questions won't be answered right away, but Burger King doesn't have much of a choice. Breakfast is big business, and chains have to stand out one way or another. Another Shot in the Battle for Breakfast It isn't easy to make a difference in the morning. Just ask Wendy's (WEN), which has already retreated twice from the national breakfast market over the past decade. The first time it thought that it could set itself apart by offering sausage gravy-soaked biscuits and breakfast versions of its then-popular Frescata sandwiches. More recently it tried to stick closer to the McDonald's (MCD) playbook with oatmeal, breakfast burritos and biscuit sandwiches. That didn't pan out either, and last year it discontinued breakfast at all but less than 10 percent of its restaurants. It's not just the burger flippers hoping to woo diners with caffeinated mornings. Subway -- the world's largest restaur

But as highlighted earlier Whitecap's Canadian light sweet crude is not as heavily discounted as Canadian heavy oil or bitumen. This does not leave it exposed to the same price risks and volatility as those companies that have a significant portion of their production made up by Canadian heavy oil and Bitumen, such as Husky Energy (HUSKF.PK), Suncor (SU), Imperial Oil (IMO) and Canadian Natural Resources (CNQ).
